There are 2 characters that can only be unlocked in Adventure Mode. One of them is Maxwell, the guy who talks to you at the start of the game. The other is Wes, a mime. You have to find and solve a puzzle to unlock Wes. When you go in to the Door, your inventory becomes empty, except that you get to keep any unique item (e.g. Willow's Lighter) and you get a Divining Rod. I have yet to beat a single Chapter, much less 5 of them.
